[ USAGE ]

Reconstitute in 20mM Tris, 150mM NaCl (pH8.0) to a concentration of 0.1-1.0mg/mL. Do not vortex.

[ STORAGE AND STABILITY ]

Storage: Avoid repeated freeze/thaw cycles. Store at 2-8°C for one month. Aliquot and store at -80°C for 12 months. 

Stability Test: The thermal stability is described by the loss rate. Thelossratewas determined by accelerated thermal degradation test, that is, incubatetheprotein at 37°C for 48h, and no obvious degradation and precipitation wereobserved. The loss rate is less than 5% within the expiration dateunderappropriate storage condition.

Vascular Endothelial Growth Factor 121 (VEGF121), a secreted glycoproteinmember of the VEGF family and a VEGFA splice variant, lacks heparin-bindingdomains for free extracellular solubility. It binds VEGFR121 on vascularendothelial cells to exert robust pro-angiogenic effects, promoting endothelial cell proliferation, migration and tube formation to drive vasculogenesisandangiogenesis. Distinct from anti-proliferative/pro-apoptotic proteins, it inhibitsendothelial cell apoptosis via survival signaling, sustaining vascular integrityandmediating tissue vascularization in development, wound repair and pathological angiogenesis.To test the effect of VEGF121 on cell apoptosis, HCT116cellswere seeded into triplicate wells of 96-well plates at a density of 5,000cells/well and allowed to attach, replaced with serum-free overnight, then the mediumwasreplaced with 5% serum standard DMEM prior to the addition of variousconcentrations of recombinant human VEGF121. After incubated for 48h, cellswere observed by inverted microscope and cell proliferation was measuredbyCell Counting Kit-8 (CCK-8). Briefly, 10 μl of CCK-8 solution was addedtoeachwell of the plate, then the absorbance at 450 nm was measuredusingamicroplate reader after incubating the plate for 1-4 hours at 37 ℃. Cell viabilitywas assessed by CCK-8(Cell Counting Kit-8 ) assay after incubationwithrecombinant VEGF121 for 48h. The result was shown in Figure 1. It was obviousthat VEGF121 significantly decreased cell viability of HCT116 cells. TheED50of recombinant human VEGF121 is 153 ng/mL.
